I have IIS7 (win2008 32bit) with https and Client Certificate Required.
I have many partners configured to post request and they are getting reply.
But one of my partner is facing issue. Sometime iss start reply with error code 500 (sometime it works) and upon investigation i can not see request is reaching to application no logs at all. In iis logs detail error code is 500 0 64. My other partner
are working fine none them having any issue.
In detail tracing it is clear that iis is responding back without sending request to application. We have enable network monitor and capture traffic between server and client but could not find anything unusual i.e ssl handshake

What was "the detail tracing"? When you are clear that 500 0 64 indicates an error, use Failed Request Tracing to analyze the exact request/response workflow. That can tell which module is responsible and then you gain better insight. Other tracing such
as Network Monitor is less useful at this stage.

Have actual problems been reported that correspond to the 500s? Or is this just something you found when perusing the logs?

The win32-status code 64 means "The specified network name is no longer available".
This link provide the solution for this issue:
It looks just like the user don't have permission to access pass through the client certificate authentication.
