IIS7 New Site - 404.17 Error

I have little exp. with IIS. I was given a folder with the website files, web config and an access db. I put the site files in a folder on a secondary volume (D:). I created a new site and pointed to the source folder. When I try to view the site I get a
"404.17 not found" error. If I go to basic settings for the site and click Test Connection I get a pass for authentication but a failure of the Authorization test. The error is "Cannot verify access to path..." with the path to the site and application.
What I've tried so far...
- I've confirmed I'm set to 32bit for the App Pool.
- I've granted the domain\computer account and NETWORK SERVICE accounts read access to the web folder.
- I've set the connect as for the site to a local admin account, which eliminated the error for the Authorization test.
I'd appreciate some help in walking through trouble shooting this further.

Thanks for the link. I looked over the document and was not able to determine my problem. If I assume that my problem is related to a handler, as the error would seem to indicate, I'm unclear where to find the configuration of that in my web.config.
I'd be happy to post my configuration if that would help.

If I do a "Connect As" for the site, and use local admin credentials, I'm able to get past the 404.17 error.
