Email Stays in Queue folder

I belive there is a genius who can tell me what i m doing wrong.
As I am a at beginner level, I tried a lot of things which i read from forums etc but couldn't solve it yet
I am working on a web app which send emails. The email stays at Queue folder in "C:\Inetpub\mailroot" I have played with 'Connectoin control' , Relay Restrictions, assigning diffent IPs, including 127.0.0.1. ETC
but nothing works. It shows me following message in Event viewer--> system

I HAVE TO FINISH IT QUIKLY , I HAVE SPENTS SO MUCH TIME ON IT. DOES REINSTALLING THE IIS WILL MAKE ANY DIFFERENCE.

The remote server did not respond to a connection attempt.
The destination is not answering. As long as you know the address is correct, and you have checked the SMTP log file for possible hints, it's a problem outside your control.
