error in folder permissions change resulting in Server Application Un...

Someone recently changed permissions of an unknown number of folders and now all .NET sites display the following when accessed through HTTP:
The web application you are attempting to access on this web server is currently unavailable. Please hit the "Refresh" button in your web browser to retry your request.
Administrator Note: An error message detailing the cause of this specific request failure can be found in the application event log of the web server. Please review this log entry to discover what caused this error to occur.
The tech doesn't remember what folders he changed, but he knows he was doing it in our root C:\ folder.
Looking around here it seems that perhaps the user group IIS_WPG got removed from some folders it needed access to. Does anyone know if this is accurate, and if so, what specific folders are important? If it could be something else, can anyone elaborate
