i have enable the IUSR_MACHINENAME in my server,but i can't log on my website,and report a 401.1 error,please help me!!!

401.1 is login failed. Not sure what you mean when you say you enabled the IUSR account, it really doesn't have anything to with this error anyway. Are you using Windows authentication? If so, you need a valid Windows account that has access. If you
don't want authentication, turn it off and enable only anonymous.

A 401.1 will happen if the IIS MMC has the wrong password for IUSR ...

When you say you have enabled IUSR_Machine name i take that you have enabled Anonymous authentication... and when anonymous authentication is enabled did you check of the same account ie. IUSR_machinename is having the proper NTFS permissions on the content.
